چکیده

As a treatment modality for malign and certain non-malignant diseases, photodynamic therapy (PDT) involves a two step protocol which consists of the (selective) uptake and accumulation of a photosensitizing agent in target cells and the subsequent irradiation with light in the visible range. Reactive oxygen species (ROS) produced during this process cause cellular damage and, depending on the treatment dose/severity of damage, lead to either cellular repair/survival, apoptotic cell death or necrosis. PDT-induced apoptosis has been focused on during the last years due to the intimate connection between ROS generation, mitochondria and apoptosis; by this PDT employs mechanisms different to those in the action of radio- and chemotherapeutics, giving rise to the chance of apoptosis induction by PDT even in cells resistant to conventional treatments. In this review, the (experimental) variables determining the cellular response after PDT and the known mechanistic details of PDT-triggered induction and execution of apoptosis are discussed. This is accompanied by a critical evaluation of wide-spread methods employed in apoptosis detection with special respect to in vitro/cell-based methodology.

APOPTOSIS INDUCTION BY PHOTODYNAMIC TREATMENT WITH ENDOGENOUS PROTOPORPHYRIN IX? Short title: Apoptosis by ALA-PDT?

v Apoptosis can be induced in several cell lines by photodynamic treatment with different photosensitizers including aminolevulinic acid (ALA)-stimulated endogenous protoporphyrin IX (PpIX). This PpIX is successfully used as tumor marker in diagnosis, and owns good sensitizing properties for tumor therapy (PDT). The aim of the study was to investigate cell death mechanisms and its consequences ...

Determination of Vascular Endothelial- and Fibroblast-Growth Factor Receptors in a Mouse Fibrosarcoma Tumor Model Following Photodynamic Therapy

The role of angiogenic molecules, like vascular endothelial growth factor (VEGF) and fibroblast growth factor (FGF) in tumor angiogenesis was well confirmed. Photodynamic therapy (PDT) action is, to very high degree, based on tumor vasculature damage. Therefore, it seemed to be important to evaluate growth factor receptors after PDT. The extent of receptor expression was studied by immuno-histo...

Caspase-2: a possible trigger of apoptosis induced in A-549 tumor cells by ZnPc photodynamic treatment.

Photodynamic therapy applied to cell cultures represents a widely accepted experimental method to investigate molecular mechanisms that lead to apoptotic cell death. In this context the subcellular localization of photosensitizers seems to be a significant factor in order to determine the apoptotic pathway that could be activated. We have characterized the experimental conditions that induce ap...

Induction of Apoptosis and Non-Apoptosis in Human Breast Cancer Cell Line (MCF-7) by Cisplatin and Caffeine

Background: Molecular targeted therapy by different cell death inducers are recently considered in cancer therapy. The aim of this study was to compare the effect of cisplatin and inositol trisphosphate kinase inhibitor (caffeine) on human breast cancer cell line (MCF-7). The pattern of cell death in MCF-7 cells following the exposure to cisplatin and caffeine in individual and combination form...

Hypericin-photodynamic therapy inhibits proliferation and induces apoptosis in human rheumatoid arthritis fibroblast-like synoviocytes cell line MH7A

Objective(s): To elucidate the effects and potential mechanisms of hypericin-photodynamic therapy (HYP-PDT) for treating the human rheumatoid arthritis (RA) fibroblast-like synoviocyte (FLS) MH7A cell-line. Materials and Methods: MH7A cells were subjected to HYP-PDT intervention and apoptosis was evaluated via MTT, nuclear staining, and flowcytometry analyses. Intracellular reactive oxygen spec...
